Systematic Mapping of Bacterial CRISPRa Systems for Synergistic Gene Activation Reveals Antagonistic Effects

Abstract

This study systematically characterizes targeting rules for several bacterial CRISPR activation systems in a shared promoter context. Testing combinations at their optimal target positions did not produce synergistic activation and frequently revealed antagonistic effects, highlighting important mechanistic differences between bacterial and eukaryotic transcriptional activation.
