CRISPRi-assisted E. coli strains increase success rate of burdensome construct cloning

Abstract

This preprint presents CRISPR interference-assisted cloning strains that reduce expression from common promoters and ribosome-binding sites while burdensome constructs are assembled in E. coli. Genome-integrated CRISPR components and customizable guide RNAs provide a practical route for improving difficult cloning workflows before constructs are transferred to their intended hosts.
